The interleukin 4 (IL4, IL-4) is a cytokine that induces differentiation of naive helper T cells (T h 0 cells) to T h 2 cells. Upon activation by IL-4, T h 2 cells subsequently produce additional IL-4 in a positive feedback loop.

STAT6 is so vital for the actions of IL-4 that Stat6-deficient mice phenocopy many of the defects observed with Il4- or Il4ra-deficient mice (e.g., related to Th2 differentiation and Ig class switch).

IL-4 plays important roles in a myriad of cellular events, making it difficult to study its effects, particularly in vivo, on any one system. Although this review focuses on its neurological functions, most of what is known about IL4 comes from studies of peripheral immune cells, such as macrophages and lymphocytes.
