There is a group of guys that have done the 125cc but haven't seen any 250cc. The engine vibration would be pretty bad I would imagine and the RS50 frame not sure can cope with the engine output. You would be better off finding a RS125 rolling chassis cheap then that frame would be fine. I know of a guy that did a RGV250 engine into a RS125 ...

In terms of quality, the Derbi cannot hold a candle to the RS50. Look at the two bikes side by side and compare the plastics used, the quality of the various fasteners (screws, bolts and so on) and even the instrumentation. The RS50 is the better bike in that respect, it's made from far better and far more expensive materials.
