2018年3月29日 · Loss of XIAP in mouse and human cells inhibited starvation-induced degradation of LC3 proteins and an autophagy substrate p62. It also led to the accumulation of mature autophagosomes, suggesting that XIAP controls autophagic flux by mediating autolysosome formation.

The p62 cargo receptor has a major function during the degradation of misfolded, ubiquitinated proteins by selective autophagy; here it functions to phase separate these proteins into larger condensates and tether them to the autophagosomal membrane.

XIAP functioned as an ubiquitination E3 ligase towards p62 and suppressed p62 expression through ubiquitin-proteasomal degradation. Furthermore, XIAP enhanced cancer cell proliferation, viability, and colony formation in vitro via suppression of p62.

2018年1月9日 · Our study deciphered that IAp induced apoptosis and autophagy through p62-dependent oxidative stress in breast cancer T-47D cells. These findings add a further piece to the jigsaw puzzle of IAp mode of action against breast cancer cell lines which will assist scientists in their attempt to develop this alkaloid into a therapeutic agent.
